Meanwhile, easyJet has started serving Almeria in Spain from Liverpool airport with flights on Tuesdays and Saturdays.
A new easyJet route from Southampton airport to Barcelona will also operate twice-weekly.
The Wizz Air flights to the two Italian cites are operating three times a week using new-generation Airbus A321neo aircraft.
The new Edinburgh-Northern Ireland connection is being operated five times a week on Mondays, Wednesdays, Thursdays, Fridays and Sundays.
The new route is the eighth service easyJet has added to its Belfast City network since starting operations there in 2021.
Glasgow airport managing director Mark Beveridge said: "We are delighted to see Wizz Air’s new services to Rome and Milan take flight from Glasgow Airport.
“These exciting additions mark a significant milestone in our partnership with Wizz Air and further enhance our connectivity to two of Europe’s most iconic cities.
“The launch of these routes not only offers our passengers greater choice and flexibility, but also strengthens Glasgow’s position as a gateway to Europe.
“We look forward to continuing our work with Wizz Air as they expand their presence in Scotland and offer even more opportunities for our passengers to explore the world.”
Belfast City airport aviation development manager Ellie McGimpsey said: “EasyJet’s new Edinburgh service further strengthens our domestic offering and provides passengers with even more choice.
“Launching ahead of Christmas, this route is perfectly timed for those planning a festive trip, while providing more opportunities for short breaks, family visits, or seasonal travel.”
